The purification and properties of superoxide dismutase from Neurospora crassa.

چکیده
Soluble extracts of Neurospora crassa contain a single, electrophoretically distinct, superoxide dismutase. This enzyme has been isolated and has been found to be a bluegreen, copperand zinc-containing enzyme, similar to that already described from bovine tissues and from garden peas. The molecular weight was ,approximately 31,000, and the enzyme appeared to be composed of 2 subunits of equal size joined only by noncovalent interractions. The Neurospora enzyme contains two Cu++ and two Zn++ per molecule. The ultraviolet absorption spectrum indicates a lack of tryptophan. Amino acid analyses are reported as are the spectral and catalytic properties.

Primary structure of copper-zinc superoxide dismutase from Neurospora crassa.
The complete amino acid sequence of copper-zinc superoxide dismutase from Neurospora crassa is reported. The subunit consists of 153 amino acids and has a Mr of 15,850. The primary structure was determined by automated and manual sequence analysis of peptides obtained by digestions of the carboxymethylated and aminoethylated enzyme with trypsin and thermolysin. The protein is devoid of tryptoph...

1. Neurospora crassa was found to contain two distinct superoxide dismutases. 2. Most of the activity is associated with the cytosolic fraction and was shown to be the Cu/Zn-containing form of the protein. 3. Mitochondria isolated from Neurospora crassa showed two distinct superoxide dismutases: a cyanide-sensitive Cu/Zn-containing protein and a cyanide-insensitive form which probably contains ...
متن کاملSuperoxide dismutases in mitochondria from Helianthls tuberosus and Neurospora crassa.
Superoxide dismutase has been studied in a wide range of organisms (Fridovich, 1975). Early observations suggested that superoxide dismutases isolated from eukaryotes (McCord & Fridovich, 1969; Misra & Fridovich, 1972; Beauchamp & Fridovich, 1973) showed little diversity in their properties, having mol.wts. near 32000 and containing two copper and two zinc atoms per molecule. The activity of th...
متن کاملSuperoxide dismutase (sod-1) null mutants of Neurospora crassa: oxidative stress sensitivity, spontaneous mutation rate and response to mutagens.
Enzymatic superoxide-dismutase activity is believed to be important in defense against the toxic effects of superoxide. Although superoxide dismutases are among the best studied proteins, numerous questions remain concerning the specific biological roles of the various superoxide-dismutase types. In part, this is because the proposed damaging effects of superoxide are manifold, ranging from ina...
متن کاملBiotransformation of Hydrocortisone by Neurospora crassa
The ability of Neurospora crassa FGSC 4335 in the biotransformation of hydrocortisone was investigated. The microorganism produced two major metabolites after incubation with the substrate for seven days. Each microbial product was purified chromatographically and identified on the basis of spectral data. The products were identified as 11?,17?,20?,21-tetrahydroxypregn-4-en-3-one (II) and 11?-h...
